Sample Collection Instructions
Thank you for choosing Flock Forensics! Please read these instructions carefully before collecting your sample. Proper collection helps ensure accurate DNA results and prevents delays.
Materials Needed
You may use your own supplies or purchase a Flock Forensics Sample Collection Kit.
Recommended Supplies
- Nail clippers
- Alcohol wipes
- Disposable gloves
- PCR tube, coffee filter strip, clean paper, or cotton swab (Q-tip)
- Small resealable plastic bags
- Bird leg bands or another method to identify each bird
- Permanent marker or pen
- QuikClot or styptic powder (optional, but recommended)
Before You Begin
- Work in a clean, well-lit area.
- Wash your hands or wear disposable gloves.
- Clean all tools before each bird to prevent cross-contamination.
- Use alcohol to disinfect clippers or the bird's toe, allow everything to dry completely before collecting the sample. Wet alcohol can interfere with lab testing.
- Label samples for accuracy.
- Never allow samples from different birds to touch each other.
- Each bird must have its own collection material and its own labeled bag.
Option 1: Blood Sample (Recommended)
Blood provides the highest quality DNA and is recommended whenever possible.
Step 1
Gently but securely hold your bird.
Step 2
Clean one toenail with an alcohol wipe and allow it to dry completely.
Step 3
Using clean clippers, trim just the tip of the toenail, slightly below the quick. The quick usually appears as a cloudy or pinkish area inside the nail.
Step 4
Allow a small drop of blood to form.
Collect the blood by touching it to the inside of a PCR tube lid, clean coffee filter strip, clean paper, or cotton swab. If needed, gently massage the toe downward to encourage another drop.
Only a small visible amount of blood is needed. More blood is not better. Excess blood in PCR tubes can cause spoilage. Dry blood will have a longer shelf life and is recommended.
Step 5 (Optional)
Apply QuikClot or styptic powder to stop the bleeding.
Option 2: Feather Sample
Feathers must contain the quill / tissue at the base of the shaft to provide usable DNA.
- Collect 4-6 freshly plucked chest or body feathers. Pull feathers in the direction of growth to avoid breakage.
- Do not use naturally shed feathers or chick "fluff".
- The soft tissue attached to the bottom of the feather (the quill) is what contains the DNA.
- Allow any blood on the feather to dry before packaging.
- Place feathers from each bird into their own labeled plastic bag.
Option 3: Egg Membrane Sample
Membrane samples work best when collected from newly hatched chicks from small hatches.
Step 1
After the chick hatches, locate the inner membrane still attached to the shell.
Step 2
Using clean hands or gloves, remove the membrane from the shell.
Step 3
Allow the membrane to air dry completely.
Step 4
Place the dried membrane into its own labeled plastic bag.
Packaging Your Samples
- Each bird must be packaged separately.
- Use a separate labeled bag for every sample.
- Clearly label each bag with the bird's identifier.
- Do not use the same identifier for multiple birds in the same order.
- Ensure all non-blood samples are completely dry before sealing.
